Dna from Herbarium Specimens: Ii. Correlation of Dna Degradation with Humidity
نویسنده
چکیده
Comparisons were made of DNA extracted from leaves of Juniperus virginiana stored in 100, 75, 70, 54 and 23% humidity at 21oC for up to 12 months. Fungal growth was observed in the 100, 75 and 70% humidity tests that resulted in the degradation of the juniper DNA. The DNA in leaves stored at 54.4 and 23.1% RH show no evidence of degradation after 12 months storage. It appears that storage of herbarium specimens at sub-ambient temperatures and at RH less than 55% should be effective in preserving DNA in situ. Phytologia 92(3): 351-359 (December 1, 2010).

Comparison study of three methods for genomic DNA extraction from fresh and herbarium leaf specimens of Achillea wilhelmsii C.Koch
DNA extraction from plant tissues often causes most problems. For example, unsuccessful removal secondary metabolites during extraction, such as phenolic compounds in aromatic and medicinal plants, cause to some mistakes in result of molecular experiments by using of the extracted DNA. Achillea wilhelmsii is a medicinal plant belong to Asteraceae family and native to Iran, there is little infor...
متن کاملTemporal patterns of damage and decay kinetics of DNA retrieved from plant herbarium specimens
Herbaria archive a record of changes of worldwide plant biodiversity harbouring millions of specimens that contain DNA suitable for genome sequencing. To profit from this resource, it is fundamental to understand in detail the process of DNA degradation in herbarium specimens. We investigated patterns of DNA fragmentation and nucleotide misincorporation by analysing 86 herbarium samples spannin...
متن کاملDNA Damage in Plant Herbarium Tissue
Dried plant herbarium specimens are potentially a valuable source of DNA. Efforts to obtain genetic information from this source are often hindered by an inability to obtain amplifiable DNA as herbarium DNA is typically highly degraded. DNA post-mortem damage may not only reduce the number of amplifiable template molecules, but may also lead to the generation of erroneous sequence information. ...
متن کاملA non-destructive DNA sampling technique for herbarium specimens
Herbarium specimens are an important source of DNA for plant research but current sampling methods require the removal of material for DNA extraction. This is undesirable for irreplaceable specimens such as rare species or type material. Here I present the first non-destructive sampling method for extracting DNA from herbarium specimens. DNA was successfully retrieved from robust leaves and/or ...
متن کاملHow to Open the Treasure Chest? Optimising DNA Extraction from Herbarium Specimens
Herbarium collections are potentially an enormous resource for DNA studies, but the use of herbarium specimens in molecular studies has thus far been slowed down by difficulty in obtaining amplifiable DNA. Here we compare a set of commercially available DNA extraction protocols and their performance in terms of DNA purity and yield, and PCR amplification success as measured by using three diffe...
